Urea based foldamers

چکیده
N,N?-linked oligoureas are a class of enantiopure, sequence-defined peptidomimetic oligomers without amino acids that form well-defined and predictable helical structures akin to the peptide ?-helix. Oligourea-based foldamers combine number features—such as synthetic accessibility, sequence modularity, folding fidelity—that bode well for their use in range applications from medicinal chemistry catalysis. Moreover, it was recently recognized this backbone can be combined with regular peptides generate helically folded peptide-oligourea hybrids display additional features terms helix mimicry protein-surface recognition properties. Here we provide detailed protocols preparation requested monomers synthesis purification homo-oligoureas hybrids.

Replica Exchange Molecular Dynamics Modeling of Foldamers
Introduction Oligo(m-phenyleneethynylene)s, shown in Figure 1, have been shown to undergo a solvent-driven transition from a random conformation in low polarity solvents to an α-helix conformation in polar solvents as a result of solvophobic interactions. 2 However, a detailed atomic-level study of the effects of specific intermolecular interactions, such as hydrogen bonding, van der Waals and ...
متن کاملAntibacterial mechanism of action of arylamide foldamers.
Small arylamide foldamers designed to mimic the amphiphilic nature of antimicrobial peptides (AMPs) have shown potent bactericidal activity against both Gram-negative and Gram-positive strains without many of the drawbacks of natural AMPs. These foldamers were shown to cause large changes in the permeability of the outer membrane of Escherichia coli. They cause more limited permeabilization of ...
متن کاملFoldamers: from Design to Protein Recognition
Proteins and nucleic acids perform a wide range of complex functions in biological systems. Nearly all of these molecular operations require the biopolymer chain to adopt a compact and specific folding pattern. The conformational behavior of biopolymers is usually analyzed hierarchically: secondary structure reflects local features of the backbone (helix and sheet are the secondary structures w...
متن کاملA post-modification approach to peptide foldamers.
Hydrophilic/hydrophobic patterning is a powerful strategy to control folding in non-natural polymers/oligomers. In this contribution, we present a novel strategy for the preparation of alternating hydrophilic/hydrophobic patterned non-natural peptide foldamers. This strategy relies on the post-modification of a reactive peptide precursor that can be prepared via standard solid phase peptide syn...
متن کاملHow did prebiotic polymers become informational foldamers?
A mystery about the origins of life is which molecular structures – and what spontaneous processes – drove the autocatalytic transition from simple chemistry to biology? Using the HP lattice model of polymer sequence spaces leads to the prediction that random sequences of hydrophobic (H) and polar (P ) monomers can collapse into relatively compact structures, exposing hydrophobic surfaces, acti...
